Abstract Objective To establish a method for the determination of compound sulfur cream. Methods UV spectrophotometry was used to directly determine the content of sublimed sulfur in the cream. The measurement wavelength was 281.6 nm, and the average recovery of sublimed sulfur was 99.5%, and the RSD was 0.99%. Conclusion This method is simple, rapid and accurate, and is suitable for hospital preparation analysis. Abstract Objective To establish a method for determination compound sulfur cream. Key words UV spectrophotometry Sublimed Sulfur Cream Compound sulfur cream is a commonly used preparation for hospital dermatology, mainly for the treatment of acne, acne, rosacea and other symptoms. The prescription is: sublimation sulfur lOOg, camphor l0g, menthol l0g, matrix added to 1000g (Changzhou Hospital Preparation Regulations 1986: 160). The quality control of this preparation has only been used as an identification test in the past. In order to improve the detection standard, the content of the preparation is now measured. The literature method for determining the sulfur content in ointment preparations is cumbersome and time consuming. The solvent was used as a blank and scanned automatically in the wavelength range of 240 nm to 400 nm to obtain an ultraviolet absorption spectrum. It can be seen that the matrix also absorbs at the wavelength of 264.0 nm of the sublimation sulfur absorption peak, while the matrix has no absorption at the wavelength of the shoulder peak of 281.6 nm, so 281.6 nm is selected as the measurement wavelength to eliminate the interference of the matrix. 2.2 Drawing of the standard curve: Weigh accurately 0.05073g of sublimed sulfur dried in a phosphorus pentoxide dryer for 4h, placed in a 50ml volumetric flask, dissolved in the solvent, diluted to the mark, and shaken. Accurately draw 0.20, 0.30, 0.40, 0.50, 0.60, O7ml in a 25m1 volumetric flask and dilute to volume. A standard series solution containing sublimed sulfur of 8.12-28.41 μg/ml was prepared, and the absorption value was measured at 281.6 nm with the solvent as a blank. The absorption value A and the concentration C were regressed, and the regression equation was obtained: C=39.7182A+0.987 r=O.9998(n=6) 2.3 Recovery test: Accurately weigh 5.12552g of sublimed sulfur, make 50g of cream according to the prescription process, accurately weigh 1g of cream into the beaker, add solvent about 90ml, stir After dissolving, transfer to a lO0 ml volumetric flask and dilute to volume with a solvent. Accurately draw 5ml to 25ml volumetric flask and dilute with solvent. Accurately digest 1.6, 2.0, 2.4 ml each of 3 parts in a 25 ml volumetric flask and dilute with a solvent. Taking the solvent as a blank, the absorption value was measured at 281.6 nm, substituted into the regression equation, and C and recovery were determined according to the regression equation. 2.4 Stability test: Take a sample and prepare the measurement solution according to the method of recovery test. At room temperature and in the dark, the measurement is performed once every 1h (the absorption pool is covered), and the absorption value is basically unchanged within 4 hours. 2.5 Repeatability test: 6 parts of the same sample were accurately taken and determined according to the method of recovery test. The result was X. 101.7% and RSD was 0.43%. 2.6 Sample determination: Take 3 batches of different batches of cream, accurately weigh 1g, and determine the content of sublimed sulfur according to the method of recovery test. The content of sublimed sulfur is in the range of 90.0%/~110.0% of the labeled amount. 3 Discussion 3.1 According to the solubility properties of the main drug and auxiliary materials, the experimental selection - absolute ethanol (1:1) as the solvent, prepared according to the experimental method, can produce a clear, colorless and transparent measuring solution. Moreover, anhydrous ethanol has no ultraviolet absorption at a wavelength greater than 250 nm and does not interfere with the measurement. 3.2 The substrate in the prescription is: monostearyl 50g, stearic acid lOOg, white petrolatum 5og, liquid paraffin 150g, glycerol lOOg, sodium lauryl sulfate, triethanolamine 2ml, paraben ethyl ester 1g, distilled water added to lO00g. 3.3 In the scanning wavelength range, the assay liquid of camphor and menthol has no UV absorption and does not interfere with the determination of content. 3.4 This method directly determines the content of sublimed sulfur in the cream without separation. The method is simple, rapid and accurate, and is suitable for the analysis of the pharmaceutical preparation.
